5.1 Culture medium

RPMI 1640+20% FBS

5.3 Cell passaging

5.3.1 The culture process can be maintained by adding fresh culture medium. Or the cell density can be resuspended by centrifugation to reach 2×104~4×10^4/ml.

5.4 Tips for electroporation

5.4.1 Electrotransfer buffer: EK buffer

5.4.2 For each electroporation, add 1 μg of plasmid into 100 μl electroporation volume. The concentration of plasmid extraction is 1.0-2 μg/ul (A260/280=1.8-2.0). System: 3.0~5.0×10^5 cells/60 μl electroporation buffer/well.
