5. Culture Conditions

5.1 Culture medium

DMEM+10% FBS

5.3 Cell passaging

5.3.1 During the digestion process, do not blow or shake the culture flask to prevent cell clumping.

5.3.2 Place in a 37℃ incubator for digestion.

5.3.3 Subculture at a 1:3 ratio every other day.

5.4Transfection tips

5.4.1 Electrotransfer buffer: EK buffer

5.4.2 For each electroporation, add 1 μg of plasmid into 100 μl electroporation volume. The concentration of plasmid extraction is 1-2 μg/ul (A260/280=1.8-2.0). System: 1.5~5.0×10^5 cells/60 μl electroporation buffer/well.
